John Clarke was born on 16 August 1823.1 He was the son of Sir Charles Mansfield Clarke, 1st Bt. and Mary Anna Squire.2 He married Helen Alexande, daughter of Henry Alexande, on 30 January 1855.1 He died on 8 May 1906 at age 82.1
He graduated with a Doctor of Medicine (M.D.)1 He was appointed Fellow, Royal College of Physicians, London (F.R.C.P.)1
